Background:

A client with an anti-infectives program had six chemical series of compounds with excellent efficacy profiles and wanted information for selection of a chemical series and a backup for preclinical studies.

Problem:

• Nothing was known about ADMET toxicology properties of the compounds.

Solution:

• Apredica scientists performed studies that focused on toxicity of a representative compound of each series.
• A mammalian cell toxicity assay was performed as a general mammalian screen.
• A hERG/Cyp450 screen was performed to identify any cardiac toxicity and drug-drug interaction potential early.

Results:

• Series B and D had the safest profiles; series E and F were eliminated.
• The client was able to prioritize the compounds for preclinical testing.
