Safety assessment prior to human clinical trials has become the norm in drug development. It is crucial for reducing attrition in phase 2 and later development, where 75% of failures is now due to safety and metabolism issues. To address this hurdle, the drug discovery and development process is undergoing a paradigm shift. Discovery and development teams are functionally merging so that safety issues are investigated much earlier than ever before.

The most common safety issues are associated with CYP450 and ion channels. CYP450 inhibition can exacerbate interaction with ion channels. A prominent recent example of the consequences of failure to investigate cardiac risk and CYP-3A4 inhibition is the withdrawal of Terfenadine from the market.

This seminar will feature:

  • How cardiac safety and other ADME and toxicity issues can be assessed early, quickly, and economically.
  • Case studies of drugs withdrawn from the market due to cardiac safety.
  • How to use hERG data.
  • Literature review on the correlation of human in vivo QT prolongation with available in vitro tests.
  • Strategies and techniques for early assessment of cardiac risk.
  • Which therapeutic areas and types of drug-discovery programs should be most concerned about cardiac safety.
  • Ask the experts your questions about how to address cardiac safety issues in your programs.

Speakers:
Arthur Brown, M.D., ChanTest
Dr. Arthur M. "Buzz" Brown is the founder and CEO of ChanTest, a drug safety testing company specializing in ion channels. He has more than 30 years of experience in ion channel biophysics. His research delineated the pores of voltage-gated and inwardly rectifying potassium channels. At present, his research is on ion channel trafficking with special emphasis on the hERG channel. He holds seven patents concerning ion channels and prevention and treatment of cardiac arrhythmias. He is Adjunct Professor of Physiology and Biophysics, School of Medicine, Case Western Reserve University. Dr. Brown has organized two academic departments at the University of Texas Medical Branch and Baylor College of Medicine and one research institute, the Rammelkamp Center for Education and Research, MetroHealth Campus, Case Western Reserve University.
