Many of the drugs removed from the market have been found to be mitochondrial toxicants. The cytotoxicity of drug candidates is frequently determined using cultured cell lines, which may not be sensitive to mitochondrial toxicants. Apredica grows cells in medium that is specially formulated to optimize sensitivity to mitochondrial toxicants (MITO+) and in medium in which the cells are resistant (MITO-) to mitochondrial toxicants. Cytotoxicity of compounds is assessed by titration with both cell lines. MITO+ cells are more sensitive to mitochondrial toxicants (Toxicant) than MITO- cells; there is no difference in sensitivity to non-mitochondrial toxicants (Non-Toxicant).
