Monoamine Oxidase Assay for Drug Discovery
Monoamine oxidases (MAOs) are located at the outer membranes of mitochondria in
numerous tissues including the liver, intestinal mucosa, and nerves. MAOs play a major
role in inactivating neurotransmitters1 . Identifying small molecules
that may inhibit MAOs at an early stage in drug discovery is critical in limiting costs associated with these small molecules.
Apredica measures inhibition of MAOs using LC/MS analysis of the product, 4-hydroxyquinoline (4-HQ), formed by MAOs from their substrate kynuramine.
Principle of the Assay
Test agent is incubated in a plate-based assay with a recombinant MAO and kynuramine. 4-HQ is analyzed by LC/MS.

MAO-A and MAO-B.
Sample Requirements
1-2 mg of solid material or DMSO stock solution.
Assay Modes
- Titration: Eight serial dilutions of test agent are tested in duplicate to determine an IC50.
- Screening: Test agent is tested in duplicate at two concentrations to determine % inhibition.
